Acanthamoeba stain: (088-0012)

Clinical Indication:

 Detection of Acanthamoeba cysts and trophozoites in ocular fluid, tissue and scrapings

Turnaround Time:

 Test is performed daily

Specimen Requirements: Patient Preparation: Withdraw vitreous humor fluid by aseptic technique. Corneal tissue or scrapings may be collected also.

Collect: Place one to two drops of fluid on labeled microscope slides. Place fluid in Page's solution for culture, if requested. Place tissue or scrapings in sterile tube for transport.

Specimen Preparation: Let slides air-dry. Submit slides and samples as soon after collection as possible.

Storage/Transport: Ambient temperature (Do not refrigerate.)

Stability: 72 hours

Causes for Rejection: Insufficient sample for slide preparation, improperly labeled samples, refrigerated Page's solution or sample (not slide)

Clinical Information:

Clearly state request for Acanthamoeba stain on lab order.

Reference Range:

No cysts or trophs observed; fungal elements will be reported if present

Note:

 Sensitivity of test is variable.

When ordering tests for which Medicare or Medicaid reimbursement will be sought, physicians should only order tests that are medically necessary for the diagnosis or treatment of the patient.  Components of the organ or disease panels may be ordered individually.  The diagnostic information must substantiate all tests ordered and must be in the form of an ICD-9 code or its verbal equivalent.
