Research Interests

Cellular Mechanisms of Rickettsial Diseases

A major goal of my research is to study the cellular mechanisms of rickettsial diseases at the ultrastructural level. The interrelationships of Rickettsia conorii and R. typhi with different host cells were studied in the recently developed mouse models of these rickettsioses, and also of an ehrlichia (IOE), closely related to Ehrlichia chafeensis, in a mouse model of fatal human monocytotropic ehrlichiosis. Ultrastructural characteristics of 15 strains and isolates of ehrlichiae belonging to three genogroups of genetically related organisms united in genera Ehrlichia, Anaplasma, Neorickettsia and a strain of Wolbachia pipientis which represents a fourth genogroup in this cluster of species in the family Rickettsiaceae, were studied in continuous cell cultures or in vivo. Differences in the structure of intracytoplasmic micro-colonies (morulae) of ehrlichiae and their interrelations with the host cells allowed differentiation of the genogroups. In immunoelectron microscopy which allows to visualize the intracellular localization of the protein molecules it was revealed that gp120 protein of E. chaffeensis is expressed only at the surface of dense-cored cells (DC), not reticulate cells (RC), and is abundantly shed into the morula cavity. This protein serves as an adhesin of DC. Iron-binding Fbp of E. chaffeensis and E. canis was found primarily in the periplasmic space of their RC and also on the intramorular fibrils and on the morula membrane but only in the morulae containing DC.

I am also interested in studying the cellular mechanisms of infection with other intracellular pathogens-bacteria, viruses and protozoans and collaborate with other researchers on campus, nationwide, and internationally on the projects involving different techniques of electron microscopy. We characterized the ultrastructure of a novel alphavirus, an arenavirus, and two rhabdoviruses, a monkeypox virus in experimentally infected ground squirrels, described the morphology of a novel chlamydia pathogenic for fish, Piscichlamydia salmonis, gen.nov., sp.nov., diagnosed cases of cutaneous leishmaniasis and amebic encephalitis caused by Sappinia diploidea.
